Wolff<br \/>\n(Professor ved K2 (bistilling) og KG Jebsen senter for autoimmune sykdommer, UiB og forskar, Haukeland universitetssjukehus (hovudstilling))<\/p>\n<p><strong>Kva jobbar eg med?<\/strong><br \/>\nEg er utdanna molekyl\u00e6rbiolog med dr.grad fr\u00e5 2005 og har hatt mange roller i forskingsgruppe G1 (Endokrin medisin) opp gjennom \u00e5ra. Eg likar meg best \u00abp\u00e5 labben\u00bb men diverre s\u00e5 har eg ikkje s\u00e5 mykje tid lengre til det. Eg pr\u00f8var likevel \u00e5 utf\u00f8re laboppg\u00e5ver sj\u00f8lv som td. har med introdusering og optimalisering av metoder \u00e5 gjere, og eg pr\u00f8var \u00e5 vere litt p\u00e5 laboratoriet kvar veke. Det meste av tida g\u00e5r til rettleiing for master-, PhD-studentar, og post.docs, og til \u00e5 koordinere og diskutere kommande og p\u00e5g\u00e5ande prosjekt og resultat med mine interne kollegaer og eksterne samarbeidspartar. Sj\u00f8lvsagt skal det ogs\u00e5 skrivast s\u00f8knadar, artiklar, rapportar etc. Det blir ein del zoom-m\u00f8ter, b\u00e5de i og utanfor corona-tid.<br \/>\nForskinga mi \u00abi eit stort perspektiv\u00bb fokuserer seg p\u00e5 \u00e5 finne \u00e5rsaksmekanismar til endokrin autoimmunitet, og vi ser meir og meir p\u00e5 korleis vi kan nytte den kunnskapen til \u00e5 betre terapi for slike sjukdommar. Meir detaljert s\u00e5 leiar eg no to prosjekt, der det eine har som m\u00e5l \u00e5 identifisere monogene autoimmune sjukdommar ved \u00e5 genomsekvensere familiar med opphopning av slike tilstandar, og det andre skal \u00absingle cell\u00bb analysere regulatoriske T celler hos pasientar med autoimmune sjukdommar for \u00e5 finne ut korleis vi kan bruke slike celler som m\u00e5l eller verkt\u00f8y for terapi. I begge prosjekta blir det krevd samarbeid mellom ulike profesjonar for \u00e5 komme i m\u00e5l; legar til \u00e5 rekruttere pasientar, molekyl\u00e6rbiologar til \u00e5 utf\u00f8re laboratoriefors\u00f8k, og bioinformatikarar og genetikarar til \u00e5 hjelpe til \u00e5 f\u00e5 ut resultat ut av \u00abbig data\u00bb fors\u00f8ka. Etter det m\u00e5 igjen samarbeid med biologane til for \u00e5 f\u00e5 \u00abmeining ut av resultata\u00bb. Dette er krevjande, men kjempe-spennande, prosessar! Eg l\u00e6rer nye ting kvar dag!<br \/>\nTeknikkar vi utf\u00f8rer sj\u00f8lv i v\u00e5r lab er standard molekyl\u00e6rbiologi og immunologi som celledyrking, PCR, real-time-PCR, ELISA, diverse immunassays etc. Vi samarbeidar med kjernefasilitetane p\u00e5 UiB for flow cytometry, Cytof og RNA sekvensering, og med eksterne partar for td. genomsekvensering og \u00abgenome wide association\u00bb (GWAS) fors\u00f8k.<\/p>\n<p><strong>Kvifor likar eg \u00e5 jobbe p\u00e5 K2?<\/strong><br \/>\nEg kjenner ofte at eg er super-heldig som f\u00e5 jobbe med noko eg synst er spennande, og som utfordrar meg kvar dag. K2 er eit institutt som, b\u00e5de fysisk og fagmessig, ligg tett inntil Haukeland universitetssjukehus. Genar og immunsystemet har fasinert meg heilt sidan eg \u00abvisste at dei fanst\u00bb, og det \u00e5 f\u00e5 lov til \u00e5 sette seg inn i dette, og ha moglegheit til det fordi vi er s\u00e5 tett linka med medisinsk avdeling, er kjempe-g\u00f8y. Vi har mange ulike kompetansar i forskingsgruppa, og samarbeid internt er viktig b\u00e5de sosialt og fagleg. Legane i gruppa jobbar ofte ogs\u00e5 klinisk samtidig. Det gjer at vi kan finne nye problemstillingar saman med dei, som vi kan pr\u00f8ve \u00e5 l\u00f8yse p\u00e5 laboratoriet, og vegen er deretter kort til pasientane igjen. From bed-to bench-and back again. Det at vi har eit register og biobank som vi samarbeidar med sjukehuset om, er og ein fantastisk ressurs.<br \/>\nK2 og lab-blokken har veldig fine laboratoriefasilitetar, og vegen er kort til mine interne samarbeidspartar i andre forskingsgrupper (s\u00e6rleg medisinsk genetikk) og kjernefasilitetane. Kjernefasilitetane er verkeleg noko vi b\u00f8r satse vidare p\u00e5 slik at det ikkje sit ein forskar i kvart hj\u00f8rne med kvar sin maskin og som brukar tid p\u00e5 \u00e5 l\u00e6re seg dei same avanserte teknikkane.<br \/>\n\u00c5 jobbe p\u00e5 K2 gir meg og moglegheit til \u00e5 vere rettleiar p\u00e5 ulike niv\u00e5, noko eg likar kjempe-godt. Det utfordrar meg, og gjer at eg finn ut ting eg ikkje kan, som eg m\u00e5 sette meg inn i. After that, I have had many different roles and projects in research group G1 (Endocrine Medicine). I like it best &laquo;in the lab&raquo; but, unfortunately, I do not have so much time anymore for that. I still try to do lab tasks myself, especially when introducing and optimizing new methods, and I try to perform some lab task each week. I spend most of my time to supervise master&#8217;s, PhD students, and postdocs, and to coordinate and discuss upcoming and ongoing projects and results with my internal colleagues and external partners. Of course, applications, articles, reports, etc. must also be written. A lot of time is spent on zoom meetings, both inside and outside corona time.<br \/>\nMy research &laquo;in a large perspective&raquo; focuses on finding causal mechanisms for endocrine autoimmunity, and to translate this knowledge to improved therapy for patients with these diseases. In more detail, I lead two projects. The aim of the first project is to identify monogenic autoimmune diseases by genome sequencing of families with accumulation of endocrine autoimmunity, and the aims of the second is to &laquo;single cell&raquo; analyse regulatory T cells in patients with autoimmune diseases to investigate how we can use such cells as targets or tools for therapy. In both projects, collaboration between different professions is required; physicians to recruit patients, molecular biologists to perform laboratory experiments, and bioinformatics and geneticists to help get results out of the &laquo;big data&raquo; research. After that, collaboration with the biologists again is required to get &laquo;meaning out of the results&raquo;. These are demanding, but exciting, processes! I learn new things every day!<br \/>\nTechniques we carry out in our lab are standard molecular biology and immunology such as cell culture, PCR, real-time PCR, ELISA, various other immunoassays etc. We collaborate with the core facilities at UiB for flow cytometry, Cytof and RNA sequencing, and with external partners for e.g. genome sequencing and genome wide association (GWAS) experiments.<\/p>\n<p><strong>Why do I like working at K2?<\/strong><br \/>\nI often think that I am super-lucky who get to work with something that is exciting, and that challenges me every day. K2 is an institution that, both physically and professionally, is close to Haukeland University Hospital. Genes and the immune system have fascinated me since I &laquo;knew that they existed&raquo;, and being allowed to dig into this because we are so closely linked to the medical department, and that we have access to methodology that are needed, is very awarding. We have many different competences in the research group, and collaboration internally is important both socially and professionally. The doctors in the group often work clinically at the same time. This means that we can find new problems and hypotheses together with them, which we can try to solve in the laboratory, and the path is then short to turn to the patients again with new knowledge. From bed-to bench-and back again. The fact that we have a registry and biobank, that we can utilize as material for research, is a fantastic resource.<br \/>\nK2 and the lab block have very nice laboratory facilities, and the way is short to my internal partners in other research groups (special medical genetics) and the core facilities. The core facilities are truly something we should strive for so that we avoid researchers sitting in each corner with his\/her private\/own machine and who spend time learning the same advanced techniques.<br \/>\nWorking at K2 also gives me the opportunity to supervise students at various levels, something I like very well. It challenges me, and makes me find things I do not know, that I have to learn.
